I'm now in the cave of forgetfulness and recently returned from my first trip...it was instantly forgettable of course!
I now have a new option on connected cards to sell the location for a princley fee to e.g. the devils
My question is does this make me lose the route access myself?

No, you don't lose access to the Nadir if you sell the location. It's simply another way to gain "Closest to" some factions, in addition to some pretty nice loot they give you for the information.

However, according to my understanding you can only sell it once, so think it over well. I'm sitting on that decision at the moment myself, there's really no rush.

No. You can only sell the location to one faction, but you'll never lose the ability to enter the cave.
It also may be worth letting you know that the devils and urchins don't pay as well as the other two interested factions, so if you don't have roleplaying reasons to do otherwise, go with them.

I sold it for an RP reason (I really wanted a certain location), and then went back to being closest to the Constables. it isn't a lot of work to change your closest to, so if you have no interest in changing it, just sell for the item itself. edited by ochrasy on 6/5/2016
-- Ochrasy. Monster-Hunter. Dangerous and Watchful, favors the Constables. Robitaille. Persuasive and Shadowy, fond of the Devils. Herr Horst. Seeker of Revenge. Open to all social actions on all accounts. Preferably, send any MW-providing actions to Ochrasy.

One quick note, to put it into perspective, unless you absolutely NEED to be closest to Urchins or Devils asap, I really wouldn't sell it to them. What they give you only costs 680 pieces of scrap, what you get from the great game costs 3200 pieces of scrap. Revolutionaries is a decent prize, but it is MUCH easier to farm then the Great Game prize if you need it, but I do as little to support the insanity that is the Liberation of Night anyway. edited by Vel Farre on 6/6/2016

I’m never selling it. While I appreciate the insanely high reward, I feel that only people capable of finding the Cave are responsible enough to have access.
If I had no principles, I’d probably sell it to the Imperturbable Spymaster. Getting his reward isn’t technically difficult, but it’s extraordinarily tedious. He’ll probably use it to wipe people’s memories or torture them.
If I had to sell it to someone, I’d probably give it to the Urchins. Their reward is one of the worst, but I don’t think they have any malicious intent beyond hiding in the Cave. edited by Saklad5 on 6/6/2016

My character won't sell it either, out of love for the Revolutionary Firebrand. It is insanely tempting to sell it sometimes though, especially since he really wants that scrap of cloth you get from selling it to the devils. He's got a wedding to plan :P
